Not sure if this is planned or not:
I got a defense quest in a tile my guild wants to capture (so I have the guild fight options there) when I just clicked the quest button to get into the fight I didn't get any influence there.
Should this give influence? Would it have given influence and still fulfilled the quest if I would have attacked with a guild solo of the required difficulty?

Most definitely not. The quest is given by the Capital, so upon completing it you have completed a Capital's request and not a Guild's request.
